Spermatogenic cells in semen mainly include four types: ① Spermatogonial cells: they can be divided into three types (Ad, Ap and B). The nucleus of Ad type (dark A type) spermatogonia is round or oval, and there are many tiny chromatin particles in the nucleus, which can be deeply stained with hematoxylin. There is a pale area in the center of the nucleus, which is not stained. There are hemispherical nucleoli on the inner surface of nuclear membrane, usually 1~2, with different sizes and increased eosinophils. Vacuoles often appear in the cytoplasm, which are rich in glycogen particles and have a strong positive periodic acid reaction. Ap-type (bright A-type) nucleus is oval, with large chromatin particles in the nucleus, which is not easy to be stained by hematoxylin. There are 1~3 nucleoli in the nuclear membrane, which is eosinophilic. Obviously different from Ad cells, there are very few glycogen granules in the cytoplasm, and the periodic acid reaction is negative. The nuclei of type B spermatogonia are large and round, and the chromatin granules are lightly stained, but sometimes there are some small pieces or granules of chromatin, which are deeply stained. Some of them are often attached to the nuclear membrane, while others are attached to the nucleoli. Sugar-free protoplasm exists in cytoplasm. Ad and Ap spermatogonia are generally not easy to break away from basement membrane and fall off, and B spermatogonia are often seen in semen. Type B spermatogonia is the most sensitive cell to radiation. ② Primary spermatocytes: This type of spermatogenic stagnation often occurs at the end of the first meiotic prophase. The cell morphology of diploid synapses (homologous chromosome pairing) or late pachytene decoherence (previously separated paired homologous chromosome fragments) can be seen in semen.
