Inspection principle

Morphine is an opioid alkaloid extracted from opium. It is a pure opioid receptor agonist with powerful analgesic effect. Therefore, morphine is a narcotic analgesic in medicine. This kind of substance has euphoria at the initial stage of use, can't concentrate, will produce dreamy phenomenon, excessive use will cause acute poisoning, and it is easy to produce tolerance and addiction. Morphine is mainly excreted through the kidneys in the form of urine. Morphine prototype accounts for 2- 12%, most of which (60-80%) is morphine-glucoside. Opioid poisons (including opium, heroin, morphine and codeine) are mainly metabolized by the liver and quickly deacylated into 6- monoacetylmorphine, which is further decomposed into morphine. Therefore, the presence of morphine or its metabolites in urine and exceeding the threshold indicates the use of opioids.

The product uses antibody-antigen specific binding reaction and immune membrane chromatography technology to detect morphine in human urine by immune competitive inhibition method. That is to say, the competition between protein-labeled morphine (morphine carrier) and morphine in urine for antigen binding sites on antibodies is limited.

There is a test strip in the test cup cover, the test area (T) of the test strip is coated with morphine -BSA carrier conjugate, and colloidal gold paper containing morphine monoclonal antibody is fixed at the other end. If the urine sample contains morphine, morphine in urine will compete with morphine immobilized on nitrocellulose membrane for antigen binding site on morphine antibody labeled on colloidal gold. When the concentration of morphine in urine reaches above the threshold concentration of product design, they will first react with morphine monoclonal antibodies on colloidal gold paper and occupy all antigen binding sites of morphine antibodies, thus preventing morphine antibodies on colloidal gold from binding with morphine in the detection area on the membrane. The detection area cannot capture colloidal gold particles, and there is no red band. If there is no morphine in the urine sample or the concentration of morphine is lower than the threshold concentration, the morphine antibody on colloidal gold is not bound to morphine at all or is not saturated, so the morphine antibody on colloidal gold will bind to morphine in the detection area on the membrane, and the detection area will capture colloidal gold particles and present a red band, which is a negative result.

The quality control area (C) on the test strip is coated with goat anti-mouse IgG polyclonal antibody to indicate whether the reaction system of the kit works normally. The appearance of quality control line has nothing to do with the existence of drugs or drug metabolites. The appearance of color bands in the quality control area (C) shows that: ① the amount of samples added is sufficient; ② The sample runs normally on the test strip.

Sample requirements

Use disposable clean urine cups to collect human urine. It is recommended to use fresh urine without any special treatment. Urine samples can be stored for 2 days at 2-8℃ and 1-2 months at -20℃. Before testing, the refrigerated samples should be restored to room temperature, and the frozen samples should be completely dissolved and mixed before testing, otherwise the test results may be affected. It is suggested that the collected samples be tested in time.

1. Please read the instructions carefully before the test.

2. Restore the test cup cover, urine sample and quality control to room temperature (about 25℃). Please don't tear the package of the tool kit until it is ready. Before tearing open the package, keep the refrigerated kit at room temperature to avoid moisture absorption of the reagent.

3. Take out the kit from the sealed bag and mark the serial number of the patient or quality control on the kit.

4. Let the examinee collect urine directly in the urine test cup, and the urine sample amount should be higher than the minimum line.

5. Professionals are responsible for collecting urine cups containing urine samples and covering them tightly. If the lid is not closed properly, the sample in the kit may leak.

6. Make the two corners of the cup cover support the desktop, turn the urine test cup sideways for 15 seconds, and then put the urine test cup upright.

Within 7.3-8 minutes, observe the results on the display window of the cup cover.

8. In order to ensure the accuracy of testing, quality control products can be tested first in the testing process (new testers, new batches of products and daily testing should use quality control products for testing). Quality control products are not provided in the kit, and can be obtained from the manufacturer.

Judgment and interpretation of test results

Negative: two color bands appear, that is, a red line appears at the position of test line (T line) and quality control line (C line). This means that there are no morphine drugs in the sample.

Positive: only a red line appears at the position of the quality control line (line C), but no line appears at the detection line (line T), indicating that morphine drugs exist in the sample.

Invalid: The quality control line (line C) does not appear. In any case, a C-line should be formed, indicating that the sample was added and the operation was correct. No C-line indicates that the test result is uncertain and should be redone.

Note: A very shallow color band appears on the detection line (T-line), indicating that the morphine concentration in urine is close to the detection threshold of reagent strip, so a more accurate and specific method should be used for confirmation before making a positive conclusion.

Matters needing attention

1. During the test operation, the safety operation procedures shall be observed, and work clothes and gloves shall be worn correctly during the operation.

2. Urine specimens and all used articles should be treated as infectious articles.

3. This reagent is only used for qualitative screening, and can't determine the content of morphine in urine.

4. Each urine sample should use a new collection container and sampling straw to avoid urine sample pollution.

5. If you suspect that the urine sample is contaminated, please retest with a new urine sample.

Store the original package in the dark at 4-30℃, do not freeze it, and use it within the validity period.

Application mode

(1) Tear open the aluminum foil bag and take out the test reagent plate.

(2) Use the dropper attached to the package to suck the urine to be tested, and drop 3 drops into the sample adding hole.

(3) Read the results after 5 minutes, and the results will be invalid after 10 minutes.

Result interpretation

Positive: only C rays appeared without T rays, indicating that the content of methamphetamine and its metabolites in urine was higher than1000 ng/ml;

Negative: both C-ray and T-ray appear, indicating that the content of methamphetamine and its metabolites in urine is lower than1000 ng/ml;

Invalid: there is no C-ray, which means that the detection reagent plate is invalid, and it is recommended to test again.
