It starts from Yangzhou Northwest Ring Expressway in the north and crosses the river via Runyang Bridge to connects with Dantu Hub in the southwest of Zhenjiang, and goes south through the east span of Shanghui Town. Line S122 enters Jintan via Baodi Town in the east and Tingxianghu West in the west, passes through Nixiang West and Xuebun Town, crosses S340 and Ningchang Expressway, and continues southward through Houzhou Town and Qianma Town in Liyang to Ning Xinchang hub of Hangzhou Expressway, with a total length of 65.64 kilometers.

The Yangzhou-Liyang Expressway passes through the Runyang Bridge and connects to the north line of the bridge.
